چکیده

This finding has caused great concern with round spermatid enthusiasts. A major complaint is that “one should not restrict the label of round spermatid only to those cells in which an acrosomal vesicle is visible” (p. 2977) (4). However, our observation is based not only on phase-contrast evaluation of TESE microdroplets but also on detailed stained histology of many hundreds of testis specimens and on the previous literature on the histology and pathology of maturation arrest (5–10). The atlas of phase-contrast wet preparation cells published by Johnson et al. (5, 6) attests to the difficulty of distinguishing round spermatids from Leydig cells, Sertoli cell nuclei, and even spermatogonia without staining and whole tissue fixation. The only reason we concentrate on the albeit transient acrosomal vesicle with phase contrast is to provide an easy identification marker for wet microdroplet ICSI preparations. The absence of round spermatids in wet TESE preparations determined by phasecontrast visualization of the acrosomal vesicle in the intracytoplasmic sperm injection (ICSI) setting merely confirms what was already demonstrated in fixed, stained tissue specimens over many years (Figs. 1 and 2).

Is There a Place for Round and Elongated Spermatids Injection in Assisted Reproduction?

Spermatids are the earliest male germ cells with one set of haploid chromosomes. After experiments, mainly in rodents, the spermatid injection was introduced in human assisted reproduction to the treatment of men with non-obstructive azoospermia. Spermatid injection is a technique with particular difficulties that may negatively influence the outcome. The identification, isolation and the asses...

Fourteen babies born after round spermatid injection into human oocytes.

During the human in vitro fertilization procedure in the assisted reproductive technology, intracytoplasmic sperm injection is routinely used to inject a spermatozoon or a less mature elongating spermatid into the oocyte. In some infertile men, round spermatids (haploid male germ cells that have completed meiosis) are the most mature cells visible during testicular biopsy. The microsurgical inj...

The activity (calcium oscillator?) responsible for human oocyte activation after injection with round spermatids is associated with spermatid nuclei.

OBJECTIVE To compare the oocyte-activating ability of whole human round spermatids and their isolated nuclei. DESIGN Prospective study using sibling oocytes from patients undergoing spermatid conception and intracytoplasmic sperm injection treatment cycles. SETTING Private assisted reproduction laboratories and a university department. PATIENT(S) Couples with male infertility. INTERVENT...

Multiple pregnancies obtained by testicular spermatid injection in combination with intracytoplasmic sperm injection.

Recent studies have shown that the injection of spermatid cells into the human oocyte can result in normal fertilization, embryo development and even delivery of live, healthy offspring. In our study, 23 azoospermic cases with severe spermatogenetic defects in their testicular biopsy are presented. The serum follicle stimulating hormone (FSH) concentrations and histopathological results of thes...

Spermatid injection into human oocytes. I. Laboratory techniques and special features of zygote development.

Spermatid injection into the oocyte cytoplasm has been shown recently to yield viable human embryos developing to term after transfer to the mother. This study provides details of the laboratory techniques related to round spermatid injection (ROSI) and elongated spermatid injection (ELSI) and focuses on some special features of zygote development associated with the use of these types of sperm...

Reprogramming of Round Spermatids by the Germinal Vesicle Cytoplasm in Mice

The birthrate following round spermatid injection (ROSI) remains low in current and evidence suggests that factors in the germinal vesicle (GV) cytoplasm and certain substances in the GV such as the nucleolus might be responsible for genomic reprogramming and embryonic development. However, little is known whether the reprogramming factors in GV oocyte cytoplasm and/or nucleolus in GV are benef...
